Abstract
In this study, we aimed to identify a potential interaction of CD13, also called Aminopeptidase N (APN, EC 3.4.11.2), and the protease-activated-receptor-2 (PAR2) and to define the roles of both PAR2 and CD13 in colon cancer genesis and progression. CD13 is a membrane-bound ectopeptidase cleaving neutral or basic amino acid residues of proteins. PAR2 is a G-protein-coupled receptor, that is activated by proteases cleaving the N-terminal arginine to unmask the tethered ligand, which subsequently binds and activates the receptor. In this work, we tested a yet experimental CD13-inhibitor called IP10.C9A in vitro and in vivo. Therefore, we used 2D and 3D cell culture and a syngeneic subcutaneous tumor mouse model. Interestingly, IP10.C9A interfered with spheroid formation of murine and human colon cancer cells due to dose-dependent cytotoxic properties. In mice, the pharmacological inhibition of CD13 under IP10.C9A treatment as well as the genetic Knock-out (KO) of CD13 diminished the tumor growth rate. Against our expectations, the inhibition of PAR2 did not diminish colon cancer cell growth. All in all, IP10.C9A exhibited promising cytotoxic capacity against colon cancer cells in vitro and in vivo labeling IP10.C9A as a potential new compound in colon cancer therapy.

Die Rolle und funktionelle Interaktion von APN/CD13 und PAR2 in der Genese und Progression des Kolonkarzinoms

In dieser Studie zielten wir darauf ab, eine potentielle Interaktion zwischen CD13, auch Aminopeptidase N (APN, EC 3.4.11.2) genannt, und Protease-aktivierten Rezeptor-2 (PAR2) zu identifizieren und die Rolle von PAR2 und CD13 in der Genese und Progression des Kolonkarzinoms zu definieren. CD13 ist eine membran-gebundene Ektopeptidase, die neutrale oder basische Aminosäurereste von Proteinen spaltet. PAR2 ist ein G-protein-gekoppelter Rezeptor, der von Proteasen aktiviert wird, die das N-terminale Arginin abspalten und den angeketteten Liganden demaskieren, welcher folgend an den Rezeptor bindet und aktiviert. In dieser Arbeit testeten wir einen noch experimentellen CD13-Inhibitor IP10.C9A in vitro und in vivo. Dafür nutzten wir murine und humane 2D- und 3D-Zellkulturmodelle und ein syngenes subkutanes Tumormausmodell. Interessanterweise störte IP10.C9A die Formation von Sphäroiden in murinen und humanen Kolonkarzinomzellen aufgrund dosis-abhängiger Zytotoxizität. In Mäusen reduzierte sowohl die pharmakologische Inhibition von CD13 unter Behandlung mit IP10.C9A als auch der genetische Knock-out (KO) von CD13 die Tumorwachstumsrate. Entgegen unserer Erwartungen verringerte die Inhibition von PAR2 das Kolonkarzinomzellwachstum nicht. Zusammenfassend gesagt, weist IP10.C9A eine vielversprechende zytotoxische Kapazität gegenüber Kolonkarzinomzellen in vitro und in vivo auf. Dies kennzeichnet IP10.C9A als einen potentiellen neuen Wirkstoff für die Therapie des Kolonkarzinoms.
