Abstract

A novel target for cancer immunotherapy is VISTA which is expressed on T-cells, Antigen Presenting Cells (APCs) and tumor cells. I characterized VISTA on murine and human APCs in comparison to T-cells and VISTA-GFP transfected myeloid celllines. VISTA colocalized in all cells with vesicle markers along microtubule fibers. Additionally, VISTA was expressed in the nuclei and secreted exosomes. Immunogenic LPS activation released VISTA vesicles and increased the nuclear VISTA expression for a short duration. Storage of VISTA in vesicles allows a fast reaction on immunogenic stimuli. VISTA secretion in exosomes implies a messenger function for cell-cell communication and the nuclear VISTA an unknown transcriptional role. The overexpression of VISTA-GFP in HL-60 and THP-1 cellines led to increased adhesion and differentiation towards macrophages. These macrophages showed an increased phagocytosis. VISTA overexpression on tumor cells also increases the phagocytosis by macrophages. Despite the increased phagocytosis, VISTA is an immunoinhibitory regulator. This could be possible via decreased antigen presentation via MHC-II due to VISTA.

VISTA ist auf T-Zellen, Antigenpräsentierenden Zellen (APCs) und Tumorzellen exprimiert und soll zur Immuntherapie von Tumoren genutzt werden. Ich habe VISTA auf murinen und humanen APCs im Vergleich zu T-Zellen und VISTA-GFP transfezierten myeloiden Zelllinien charakterisiert. VISTA kolokalisierte in allen Zellen mit Vesikelmarkern. Zusätzlich war VISTA in den Zellkernen und sekretierten Exosomen exprimiert. Immunogene Stimulation setzte VISTA Vesikel frei und erhöhte die kurzfristige nukleare VISTA Expression. VISTA in Vesikeln ermöglicht eine schnelle Reaktion auf immunogene Stimuli. VISTA Sekretion in Exosome impliziert eine Botenstofffunktion zwischen Zellen und nukleares VISTA eine unbekannte transkriptionelle Rolle. Die Überexpression von VISTA-GFP in HL-60 und THP-1 Zelllinien führte zu erhöhter Adhäsion und Differenzierung in Makrophagen. Sowohl diese Makrophagen als auch Tumorzellen mit VISTA Überexpression zeigten gesteigerte Phagozytose. Trotz der erhöhten Phagozytose ist VISTA ein immunoinhibitorischer Regulator. Das könnte durch reduzierte Antigenpräsentation auf MHC-II durch VISTA möglich sein.
