Abstract

Heterotrophe Bakterien des marinen Lebensraumes spielen eine wichtige Rolle in den globalen Kreisläufen von Kohlenstoff, Stickstoff und Phosphor. Das marine Bakterium Phaeobacter inhibens dient als Modellorganismus für die weltweit erfolgreiche Roseobacter-Gruppe. Phaeobacter inhibens DSM 17395 wurde hinsichtlich seiner Anpassung an unterschiedliche N:P-Verhältnisse in definiertem Seewassermedium (SWM) mittels Prozess-kontrollierten Wachstumsexperimenten in Bioreaktoren untersucht. Diese zeigten zum einen, dass unter N-Limitation, der externe Stickstoff innerhalb der ersten Stunden des Wachstums aufgenommen, intern gespeichert und über die Zeit abgebaut wird, zum anderen, dass P-limitierende Bedingungen zu vierfachen Erträgen hinsichtlich optischer Dichte, Biotrockenmassekonzentration und CO2-Produktion führen. Zur Bestimmung der NH4+-und PO43--Konzentrationen in SWM wurden zwei kolorimetrische Hochdurchsatzverfahren für den Mikroplatten-Leser entwickelt und etabliert.

Marine heterotrophic bacteria contribute to global primary production by playing a key role in the cycling of the nutrients carbon, nitrogen and phosphorus. The marine bacterium Phaeobacter inhibens functions as model organism for the understanding of the success and importance of the Roseobacter-group. Phaeobacter inhibens DSM 17395 was analyzed in terms of its adaption to different N:P ratios in defined seawater media (SWM), using process-controlled growth experiments in bioreactors. It was demonstrated that N-limitation leads to a rapid uptake of external nitrogen in the first hours of growth, and building of an internal N-storage, which is degraded later on. P-limitation showed 4-fold yields concerning optical density, cellular dry weight and CO2-production. For the analysis of NH4+-and PO43--concentrations in SWM, two high throughput colorimetric assays for the microplate reader were established.
