Abstract

Die aromatischen Carbonylverbindungen Benzaldehyd, p-Anisaldehyd und Acetophenon zeigten in Kombination mit CuCl2 eine verstärkte gentoxische Wirkung auf humane Fibroblasten und isolierte DNA. Die Einzelsubstanzen wiesen keine oder nur eine sehr schwache DNA-Reaktivität auf. Bei den durch die Kombination der Carbonylverbindungen mit CuCl2 induzierten DNA-Schäden handelte es sich um Einzelstrangbrüche, Doppelstrangbrüche und Apurin- bzw. Apyrimidin-Stellen. Eine verstärkte Zytotoxizität der Kombinationen konnte anhand der akuten Toxizität, des Wachstums und der Koloniebildungsfähigkeit beobachtet werden. Für alle getesteten Kombinationen wurde eine sauerstoffunabhängige Reduktion von Cu(II) zu Cu(I) nachgewiesen. Die Bildung von Hydroxylradikalen über die Kupfer-Fenton Reaktion wurde ausgeschlossen. Für das Stoffgemisch Acetophenon/CuCl2 wurde ESR-spektroskopisch ein C-zentriertes Radikal nachgewiesen. Als ultimat DNA-schädigende Agenzien konnten reaktive Intermediate angenommen werden, die unter Beteiligung von Cu(I) und eines primär erzeugten Carboxyradikals gebildet wurden.

Combination effects of CuCl2 and benzaldehyde, p-anisaldehyde and acetophenone were determined on isolated DNA and human fibroblasts. DNA damage was negligible or absent when incubations were performed with either carbonyl compounds, or CuCl2 alone. The combinations, however, induced DNA-single strand breaks, double strand breaks and apurinic/apyrimidinic sites. Enhanced cytotoxicity measured as acute cytotoxicity, growth and colony forming ability, was also observed. The DNA-damaging reactions were characterized by the reduction of Cu(II) to CU(I), independent of molecular oxygen, and the absence of hydroxyl radical production via the copper driven Fenton reaction. A C-centered radical was detected by ESR spectroscopy in the reaction mix of acetophenone/CuCl2. It was concluded, that DNA damage occurred by reactive intermediates, formed under participation of Cu(I) and a primarily produced carboxy radical.
